There are quite a few differences. The v2 only has 1 stock aluminum block for the z axis smooth rods where the 2.1 has all aluminum bearing blocks. The new plus also has built in LCD. The new one has a few design changes but nothing major. The carriage is different between the two, the newest one also has a little bit wider x axis to allow space for dual extruders. It also comes stock with acme rods, GT2 belts and pulleys where the v2 has MXL belts and molded pulleys.
I have a plus v2 with a v2.1 carriage that has dual extruders. I have also upgraded a lot of my bearing blocks to aluminum. The v 2.1 has more bang for the buck but you can easily upgrade a v2 to match or surpass a stock v2.1. Here's mine, I have extended the y and z axis's to about 17 inches vs their stock less than 13 inch travel.
